What Is Mayzent Used For?

Mayzent contains siponimod and belongs to a group of medicines called sphingosine-1-phosphate (S1P) receptor modulators. It helps slow down the effects of multiple sclerosis by reducing nerve damage and potentially extending time without relapses.

Primary Uses

Mayzent is used for treating adult patients with secondary progressive multiple sclerosis (SPMS).

How It Works

Mayzent helps slow down MS effects such as worsening disability and decline in thinking. It may also slow down nerve damage caused by MS and increase your time without relapse.

How to Take Mayzent

Your Mayzent treatment begins with a special 5-day starter pack to gradually increase your dose, followed by your regular daily dose.

Dosage Instructions

Start with a 5-day titration pack: Day 1-2: 0.25mg (1 tablet), Day 3: 0.5mg (2 tablets), Day 4: 0.75mg (3 tablets), Day 5: 1.25mg (5 tablets). From Day 6 onwards: usual dose is 2mg once daily. Some patients with specific genotypes (CYP2C9 *2*3 or CYP2C9 *1*3) take 1mg once daily. Take at the same time each day with or without food, swallowed with half a glass of water.

What If You Miss a Dose?

Take the missed dose as soon as you remember, then continue with your regular schedule. If it’s almost time for your next dose, skip the missed dose. If you miss a dose during the first 6 days of treatment, contact your doctor – you may need to restart with a new starter pack. If you stop taking Mayzent for 4 or more days, you’ll need to restart with the starter pack.

Important: Never take a double dose to make up for a missed dose. Do not stop taking Mayzent suddenly without talking to your doctor.

Side Effects

Like all medications, this medicine can cause side effects. Most are mild and temporary, but some require medical attention.

Common Side Effects

These side effects occur relatively frequently and usually don’t require emergency care:

  • Headache
  • Dizziness
  • Diarrhea
  • Feeling unwell
  • Pain in legs or arms
  • Swollen hands, ankles, legs or feet
  • Weakness
  • Skin changes (small spots or lumps of various colors)

Seek immediate medical attention if you experience:

  • Infections (shingles, fever, sore throat, mouth ulcers)
  • Fungal or viral infections with headache, stiff neck, light sensitivity
  • Brain infection signs (weakness, visual changes, memory loss, thinking problems, walking difficulty)
  • Heart problems (slow or irregular heartbeat)
  • Skin cancer (new bumps, patches, moles, or changes to existing moles)
  • Worsening MS symptoms (muscle weakness, eye changes, tremor)
  • Difficulty breathing
  • Convulsions or uncontrolled shaking
  • Vision problems or trouble seeing colors
  • Liver problems (nausea, vomiting, abdominal pain, yellowing of skin/eyes, dark urine)

Important Warnings

Before starting Mayzent, your doctor needs to check several things about your health and run specific tests.

Who Should Not Take Mayzent

Do not use if you: are allergic to siponimod or any ingredients; have CYP2C9 *3*3 genotype; had a heart attack, angina, stroke, or certain heart failure in the last 6 months; have irregular heartbeat without a pacemaker.

Medical Conditions to Discuss

Tell your doctor if you have: infections or lowered immune system; haven’t had chickenpox or vaccination; eye or vision problems; uncontrolled high blood pressure; slow heartbeat or take heart-slowing medicines; severe breathing problems or asthma; liver problems; any other medical conditions.

Pregnancy and Breastfeeding

Avoid becoming pregnant while using Mayzent and for 10 days after stopping. Use effective birth control. Do not take Mayzent while breastfeeding. Tell your doctor immediately if you become pregnant.

Required Tests: You must have blood tests to check your genotype and complete blood count, plus an eye examination before starting Mayzent.

Drug Interactions

Several types of medicines can interfere with Mayzent and affect how it works.

  • Medicines for irregular heartbeat (quinidine, procainamide, amiodarone, sotalol)
  • Medicines that slow heartbeat (verapamil, diltiazem, ivabradine, digoxin)
  • Immune system medicines (other MS treatments, corticosteroids)
  • Live attenuated vaccines
  • Carbamazepine
  • Modafinil and fluconazole (for certain genotypes)

There are no known interactions between Mayzent and alcohol.

Driving and Operating Machinery

Be careful before driving or using machines until you know how Mayzent affects you. Mayzent may cause dizziness in some people.

Storage

Store tablets in refrigerator between 2°C-8°C. Do not freeze. After opening, only 1mg and 2mg tablets can be stored below 30°C for up to 3 months in a cool, dry place. Keep in original package and out of reach of children.

If you think you have taken too much Mayzent, seek urgent medical attention immediately. Call the Poisons Information Centre (13 11 26), contact your doctor, or go to the nearest hospital Emergency Department, even if you feel fine.
